  [Indications]

This medicine is a RAS GTPase family inhibitor indicated for adult patients with metastatic pancreatic ductal adenocarcinoma who have previously received at least one line of systemic therapy,or who are not suitable for multi‑drug combination systemic treatment.

  [Recommended Dose]

The recommended dosage is 300 mg taken orally once daily, with or without food.Treatment should continue until disease progression or unacceptable toxicity occurs.

  [Adverse Reactions]

  Most common adverse reactions(≥20%):

  rash,diarrhea,stomatitis,nausea,fatigue,vomiting,abdominal pain,edema,decreased appetite,and bleeding.

  Most common laboratory abnormalities(≥20%):

  decreased albumin,decreased calcium,decreased hemoglobin,increased aspartate aminotransferase(AST),lymphopenia,thrombocytopenia,increased alanine aminotransferase(ALT),decreased sodium,leukopenia,decreased magnesium,increased alkaline phosphatase,increased creatinine,and decreased potassium.

  [Pharmacology]

This product is a RAS GTPase inhibitor. It binds to cyclophilin A inside cells to form a binary complex;this complex then binds to activated,GTP‑bound RAS protein. The resulting ternary complex blocks the interaction between RAS and downstream effector molecules and promotes GTP hydrolysis, converting RAS to its inactive GDP‑bound state, thereby inhibiting the RAS signaling pathway.

It inhibits both wild‑type and mutant KRAS,NRAS,and HRAS,leading to suppression of tumor growth and induction of apoptosis. In RAS‑dependent pancreatic adenocarcinoma models, treatment with this agent inhibited tumor growth, induced tumor regression, and was associated with anti‑tumor immune effects.

  [Storage]

  Store at 20°C to 25°C(68°F to 77°F);excursions permitted between 15°C and 30°C(59°F and 86°F)for a short period.
