赛洛多辛(Silodosin)如何用药

Silodosin is a highly selective alpha-1A adrenergic receptor antagonist that improves urine flow by relaxing the smooth muscle of the prostate and bladder neck.

The correct dosage of Silodosin

1. Standard dose

The recommended standard dose is 8 mg each time (i.e. one 8 mg capsule), once a day.

2. Medication time

Taking it with meals can reduce the risk of adverse reactions such as dizziness and orthostatic hypotension that may be caused by the drug, and help the body absorb the drug.

3. Dose adjustment for special groups

(1) Patients with moderately impaired renal function: If the patient's creatinine clearance (CCr) is between 30-50mL/min, the dose should be adjusted to 4 mg each time (i.e. one 4 mg capsule), once a day, taken with food.

(2) Patients with severely impaired renal function: Patients with creatinine clearance (CCr) lower than 30mL/min are prohibited from using this product.

(3) Patients with impaired liver function: Patients with mild to moderate hepatic impairment do not need to adjust the dosage. However, use is prohibited in patients with severe hepatic insufficiency (Child-Pugh score ≥ 10).

4. Usage for patients with dysphagia

For patients who have difficulty swallowing capsules, you can carefully open the capsule and sprinkle the internal powder on a tablespoon of applesauce. It must be swallowed immediately (within 5 minutes) without chewing, and then drink a glass (about 240 ml) of cold water to ensure that the powder is completely taken. Do not store the mixed applesauce for later use.

Silodosin treatment course

The treatment course of Silodosin is not fixed and needs to be determined based on the patient’s condition improvement, tolerance and the doctor’s professional evaluation.

Important contraindications for Silodosin

Absolute contraindications

Patients under the following conditions are strictly prohibited from using Silodosin:

1. Those who are allergic to Silodosin or any ingredient in this product.

2. Patients with severe renal insufficiency (creatinine clearance CCr<30mL/min).

3. Patients with severe hepatic insufficiency (Child-Pugh score ≥ 10).

4. Patients who are taking strong CYP3A4 inhibitors, such as ketoconazole, itraconazole, clarithromycin, ritonavir, etc. Simultaneous use will significantly increase the plasma concentration of silodosin and cause serious adverse reactions.

Silodosin drug interactions

1. Silodosin is prohibited from being used in combination with other α-blockers (such as terazosin, doxazosin).

2. The combination of silodosin and PDE5 inhibitors (such as sildenafil, tadalafil) may increase the risk of hypotension, so be very cautious.

3. The combination of silodosin and strong P-glycoprotein inhibitors (such as cyclosporine) is not recommended.
