赛洛多辛(Silodosin)是治疗什么疾病的

Silodosin was developed by Kissei Pharmaceutical Co., Ltd. and was first launched in Japan under the brand "Urief®" in 2006. It mainly targets the symptoms of dysuria caused by benign prostatic hyperplasia (BPH).

What disease does Silodosin treat?

Benign prostatic hyperplasia (BPH)

It is suitable for the treatment of symptomatic patients with benign prostatic hyperplasia (BPH) to reduce urinary tract obstruction and relieve related symptoms (such as hesitant urination, thin urine stream, incomplete bladder emptying, frequent urination, urgency, and increased nocturia).

Silodosin can be used as an alternative to surgery, especially for patients who are waiting for surgery to correct hyperplasia or who are not suitable for surgery.

Silodosin usage and dosage

1. Route of administration

It is administered orally and needs to be taken with meals.

2. Dosage

8 mg each time, once a day.

3. Medication for special groups

(1) Patients with hepatic insufficiency: Patients with mild or moderate hepatic insufficiency (Child-Pugh Class A or B) do not need to adjust the dose.

It is not recommended for patients with severe hepatic insufficiency (Child-Pugh Class C).

(2) Patients with renal insufficiency: Patients with mild renal insufficiency (creatinine clearance 50–80 ml/min) do not need to adjust the dosage.

Patients with moderate renal insufficiency (creatinine clearance 30–50 ml/min) should reduce the dose to 4 mg once daily.

Not recommended for patients with severe renal insufficiency (creatinine clearance <30 ml/min).

Contraindications of Silodosin

1. Severe liver insufficiency (Child-Pugh Class C).

2. Severe renal insufficiency (creatinine clearance <30 ml/min).

3. It is prohibited to use this product in combination with strong CYP3A4 inhibitors (such as clarithromycin, itraconazole, ketoconazole, and ritonavir).

Special population use of silodosin

1. Pregnancy period

Pregnancy classification category B, silodosin is not suitable for female patients.

2. Lactation

Silodosin is not suitable for female patients.

3. Medication for children

Silodosin is not suitable for children.

4. Medication for the elderly

The incidence of orthostatic hypotension in patients aged ≥65 years is higher than that in young adults, but there are no other significant differences in safety and effectiveness.

5. Patients with hepatic insufficiency

The use of silodosin is not recommended for patients with severe hepatic insufficiency.

6. Patients with renal insufficiency

The use of silodosin is not recommended for patients with severe renal insufficiency. Patients with moderate renal insufficiency need to use the drug with caution and reduce the dose.
