It is suitable for the treatment of adult and pediatric patients with hepatic veno-occlusive disease (VOD), also known as sinusoidal obstruction syndrome (SOS), and renal or pulmonary dysfunction after hematopoietic stem cell transplantation (HSCT). What are the side effects of using defibrotide?

The most common side effects of defibrotide (incidence ≥10% and independent causal relationship) are hypotension, diarrhea, vomiting, nausea, and epistaxis.

The incidence of grade 3-4 reactions related to defibrotide was 7%, mainly hypotension, pulmonary and gastrointestinal bleeding, and abdominal cramps. Only 4% of patients discontinued treatment due to side effects. In other words, the side effects of defibrotide are usually mild, and patients do not need to worry too much.

During the treatment with defibrotide, the side effects are relatively mild and do not require targeted treatment. The side effects will gradually disappear as the medication time is extended. Once more serious or intolerable side effects are discovered, you must seek medical treatment in time!

It is important to note that defibrotide may enhance the pharmacodynamic activity of antithrombotic/fibrinolytic agents such as heparin or alteplase.

Concomitant use of defibrinoside with antithrombotic or fibrinolytic agents is contraindicated due to increased risk of bleeding.

Do not initiate defibrotide in patients with active bleeding. Monitor patients for signs of bleeding. If a patient bleeds while taking defibrotide, discontinue defibrotide, treat the cause, and provide supportive care until bleeding has stopped.

Precautions for defibrating sodium:

(1) Bleeding: Monitor patients for bleeding. If significant bleeding occurs, withhold or discontinue defibrotide.

(2) Hypersensitivity reaction: If severe or life-threatening allergic reaction occurs, discontinue, treat according to standard medical care, and monitor until signs and symptoms resolve.
