What are the indications for eflornithine tablets?

Eflornithine tablets (eflornithine) is an ornithine decarboxylase (ODC) inhibitor and an anti-proliferative drug. It was initially developed to treat certain parasitic diseases and has gradually shown new application potential in the field of tumor treatment. It irreversibly inhibits ODC, a key enzyme for cell proliferation, interferes with the polyamine synthesis pathway, and prevents the growth and spread of tumor cells. It has certain targeted therapeutic properties. Currently, the main indication for eflornithine is the maintenance treatment of patients with high-risk neuroblastoma (HRNB), especially for children and adults who have received standard multi-drug combination therapy and have achieved partial response to immunotherapy (especially anti-GD2 monoclonal antibodies) to reduce the risk of recurrence.

Neuroblastoma is a neuroectodermal tumor arising from the sympathetic nervous system that occurs primarily in children. High-risk neuroblastoma progresses quickly, is prone to relapse, and has complex treatments. Traditional treatments include high-intensity chemotherapy, stem cell transplantation, radiotherapy, and antibody immunotherapy. However, even after comprehensive treatment, a significant proportion of patients will still experience disease recurrence. At this time, the addition of eflornithine can be used as a new maintenance treatment option. In addition to neuroblastoma, eflornithine has use in certain non-oncology indications, such as the treatment of African trypanosomiasis (sleeping sickness). This is a fatal parasitic disease caused by trypanosome infection. Eflornithine becomes an important component of combined drug therapy by interfering with the cellular metabolism of the parasite. Especially in patients who are resistant to traditional drugs such as suramin or melarsoprol, eflornithine has shown good efficacy and safety.
