Ramosetron Hydrochloride Orally Disintegrating Tablets: A 5-HT3 Receptor Antagonist for Diarrhea-Predominant IBS
Ramosetron Hydrochloride is a selective 5-hydroxytryptamine type 3(5-HT3)receptor antagonist.Rather than simply suppressing an individual episode of diarrhea,it blocks 5-HT3 receptors to modulate gastrointestinal neural signaling,thereby affecting visceral sensation,gastrointestinal motility,and the defecation reflex.
Diarrhea-predominant irritable bowel syndrome involves multiple contributing factors,including brain–gut axis interactions,visceral hypersensitivity,altered gastrointestinal motility,and disturbances in serotonergic signaling.It is not caused solely by excessively rapid intestinal transit.When the intestinal mucosa is stimulated,enterochromaffin cells may release serotonin,which can activate 5-HT3 receptors and contribute to visceral sensory signaling and gastrointestinal motor regulation.By blocking this pathway,ramosetron may help reduce visceral hypersensitivity,slow accelerated intestinal transit,and improve stool consistency and urgency.
Because of this mechanism,5-HT3 receptor antagonists are considered a relevant therapeutic option for adults with IBS-D.In Japan,ramosetron is approved for adult IBS-D and has separate dosage recommendations for men and women.However,approved indications differ among countries and regions,formulations,and product brands.Use should always follow the locally approved product labeling and be determined by a gastroenterologist.
