Mitotane (Mitotane) is an oral chemotherapy drug, also known as the malonate ester, used to treat functional and nonfunctional adrenocortical cancers. Mitotane is best described as an adrenocytotoxic agent, and although it causes adrenal suppression, it apparently does not destroy cells. Administration of mitotane alters extra-adrenal cortisol metabolism in humans; resulting in measurable reductions in 17-hydroxycorticosteroids even though plasma levels of corticosteroids are not reduced. This drug apparently increases the formation of 6-B-hydroxycortisol. Although mitotane can target specific targets, it is classified as a chemotherapy drug because its mechanism of action is to treat cancer by inhibiting cell proliferation.
