Interactions between Pyrotinib/Areni and common drugs and precautions

Long-term use of Pyrotinib (Pyrotinib Maleate) requires careful attention to hidden side effects. Pyrotinib is mainly metabolized by the CYP3A4 enzyme. Combination with strong inducers may weaken the efficacy, and combination with strong inhibitors may increase the risk of toxicity. During medication, it is necessary to avoid using more than two drugs that prolong the QT interval at the same time, and correct hypokalemia and hypomagnesemia. When combined with capecitabine, attention should be paid to dose adjustment. The recommended dose of capecitabine is 1000mg/m² twice a day, taken within 30 minutes after a meal, and rested for 7 days for 14 consecutive days. Before taking the medication, electrocardiogram and left ventricular ejection fraction monitoring are required to ensure that the LVEF is not less than 50%. During the treatment period, the test should be reviewed every 3 cycles to prevent the risk of cardiovascular events and ensure the safety of treatment.
