【Indications】

Vigabatrin is an analog of gamma-aminobutyric acid (GABA). It can specifically bind to GABA aminotransferase and irreversibly, leading to an increase in GABA concentration in the brain, thereby exerting anti-epileptic effects. In addition to treating epilepsy, Vigabatrin can also treat infantile spasms (West syndrome, West syndrome). It is also the drug of choice for the treatment of Tsc abroad and effectively controls epilepsy.

【Usage and Dosage】

Vigabatrin is an oral drug that is rapidly absorbed after oral administration and can reach its peak blood level 2 hours after taking it. The maximum daily dose for adults is 2-3g. It is appropriate to start with 1g a day. Then, depending on the reaction after the medication, if the response is good, vigabatrin can be increased by 0.5g per week, and finally reach a daily dose of 2-3g. The recommended dose of Vigabatrin in children is 40 mg/kg/day. Continuous medication should be judged based on the patient's weight. The recommended dose for children weighing 10-15kg is 0.5-1g/day; the recommended dose for 15-30kg is 1-1.5g/day; the recommended dose for 30-50kg is 1.5-3g/day; >50kg 2-3g/day.

【Adverse reactions】

Common side effects of Vigabatrin include: fatigue/drowsiness/headache/dizziness/confusion/depression, etc. Since each patient's physical condition is not exactly the same, the degree of side effects of using vigabatrin tablets is not entirely the same. If serious side effects occur, please contact your doctor in time.

【Notes】

This drug should not be used if the patient is allergic to Vigabatrin, or if the patient is pregnant or breastfeeding. The elderly and patients with renal impairment need to consider reducing the dosage or frequency of medication. Vigabatrin is mainly digested by the kidneys.

The number of data available to investigate the use of Vigabatrin in pregnancy is limited (192 cases). 14.5% of those taking Vigabatrin during pregnancy had congenital anomalies. Among them, 64.3% were major deformities. Spontaneous miscarriage occurs in 10.9% of women taking Vigabatrin during pregnancy.

Patients taking Vigabatrin have a high incidence of visual field defects. This often occurs after several months to years of using Vigabatrin. Visual field defects can be severe, which can have real consequences for the patient. Most patients with visual field loss have no obvious symptoms.
