Silodosin is a highly selective α-1A adrenergic receptor antagonist that can improve symptoms such as dysuria, frequent urination, and urgency.

Is Silodosin available in China?

It has been launched in China and has been approved by the China National Medical Products Administration (NMPA) for the clinical treatment of benign prostatic hyperplasia. Domestic patients can obtain the drug under a doctor's prescription.

As a prescription drug, patients must be diagnosed by a professional physician and confirmed to be in compliance with the medication indications before use. Do not buy and take it by yourself.

Silodosin Indications

Indicated for the treatment of signs and symptoms of benign prostatic hyperplasia (BPH).

Silodosin usage and dosage

1. Standard dose

(1). Recommended dose: 8 mg each time (one 8 mg capsule), once a day, taken after meals.

(2) How to take: Swallow the tablet whole and take it with food (i.e. after meals) to reduce fluctuations in drug absorption and may reduce the risk of certain gastrointestinal discomforts.

For patients with difficulty swallowing, the capsule can be opened carefully, sprinkle the contents over a tablespoon of applesauce and take it immediately (within 5 minutes) without chewing, followed by drinking a glass of cool water to ensure that all the powder is taken.

2. Dose adjustment

(1) Patients with renal insufficiency

Moderate renal insufficiency (creatinine clearance CCr30-50mL/min): The dose should be reduced to 4 mg each time (one 4 mg capsule), once a day, taken after meals.

Severe renal insufficiency (CCr<30mL/min): the use of silodosin is prohibited.

(2). Patients with hepatic insufficiency

Mild to moderate hepatic insufficiency: No dosage adjustment is required.

Severe hepatic insufficiency (Child-Pugh score ≥10): the use of silodosin is prohibited.

3. Medication precautions

(1) Initial precautions: Orthostatic hypotension (dizziness when standing) may occur in the early stages of medication, especially after taking the medication for the first time. Therefore, during the initial treatment phase, you should avoid driving, operating machinery, or performing hazardous activities until you know how your body will respond to the medication.

(2) Drug interactions: It is prohibited to use it simultaneously with strong CYP3A4 inhibitors (such as ketoconazole, clarithromycin, itraconazole, ritonavir, etc.), otherwise it will significantly increase the plasma concentration of silodosin and lead to serious side effects. Concomitant use with other alpha-blockers is also not recommended.
