氯法齐明可以和贝达喹啉一起使用吗？

Currently, drug-resistant tuberculosis, especially multidrug-resistant tuberculosis, has become one of the three major challenges in tuberculosis control. Multidrug-resistant tuberculosis (MDR-TB) means that Mycobacterium tuberculosis is resistant to at least isoniazid and rifampicin. my country is one of the countries with severe MDR-TB epidemics. Many studies have shown that insufficient or inappropriate tuberculosis medication is the main cause of MDR-TB. Fortunately, the application of some new drugs such as bedaquiline, delamanid, linezolid, old drugs, and cycloserine have provided the possibility for the treatment of tuberculosis, especially the treatment of drug-resistant tuberculosis. So, can clofazimine be used with bedaquiline?

Clofazimine can be used with bedaquiline

The latest research shows that clofazimine has similar chemotherapeutic activity to rifampicin and isoniazid, and has no antagonistic effect with other anti-tuberculosis drugs. Clofazimine can inhibit the expression of DNA-dependent RNA polymerase and block the production of bacterial proteins induced by RNA, ultimately blocking and killing mycobacterial cells.

In the study, patients with rifampicin-resistant TB who received bedaquiline-containing regimens were followed to critically assess their treatment efficacy and cardiac safety. Consistent with other studies, few participants had a QT interval (QTcF) >500 ms or an absolute change from baseline (ΔQTcF) >60 ms, indicating that coadministration of bedaquiline and clofazimine is safe and that life-threatening QT prolongation is rare. This important finding led WHO to recommend that most cases of rifampicin-resistant TB be treated with a combination of bedaquiline and clofazimine.
