Pacritinib Drug Contraindications and Precautions
Pacritinib is contraindicated with strong CYP3A4 inhibitors or strong CYP3A4 inducers.Strong inhibitors can raise pacritinib exposure,while strong inducers can lower efficacy.Patients should report all prescription drugs,over-the-counter medicines,and supplements before starting treatment.
Use with caution in patients who have active bleeding.Pacritinib is usually avoided when baseline QTc is above 480 ms,and dose interruption or reduction may be needed if QT prolongation occurs.Serious active infection should generally be resolved before starting therapy.Low platelet count,arrhythmia history,hypokalemia,or hepatic/renal impairment also require evaluation.
Seek medical attention for persistent or severe diarrhea,black stools,vomiting blood,unusual bleeding or bruising,palpitations,fainting,or worsening signs of infection.Blood counts,bleeding symptoms,and ECG changes should be monitored clinically rather than managed by self-adjusting the dose
