Also called defibrinoside, it has anti-thrombotic, fibrinolytic, anti-ischemic and anti-infective effects in vivo and in vitro. It was approved for marketing in the European Union in October 2013, and was approved for marketing in the United States by the U.S. Food and Drug Administration (FDA) on March 30, 2016. It is suitable for the treatment of adult and pediatric patients with hepatic veno-occlusive disease (HVOD) and renal or pulmonary dysfunction after hematopoietic stem cell transplantation (HSCT).

One of the serious complications after hematopoietic stem cell transplantation (HSCT) is hepatic veno-occlusive disease (HVOD), also known as sinusoidal obstruction syndrome (SOS), which usually occurs 30 days after hematopoietic stem cell suppression. The incidence of HVOD after HSCT is approximately 8%-14%, and can reach 60% in high-risk patients. Seriously affect the patient's life and health. The launch of defibrinated sodium brings new options for patient treatment.

Is defibrinated sodium effective in treating hepatic veno-occlusive disease?

The therapeutic effect of defibrotide sodium has attracted the most attention after its launch. The use of heparin in the treatment of diseases such as hepatic veno-occlusion often increases the patient's risk of bleeding. However, defibrination sodium alone or in combination with heparin not only achieves a good preventive effect on HVOD, but also has a very low incidence of side effects.

A multi-center randomized phase III clinical trial was completed in Europe. The trial included 356 stem cell transplant patients. The incidence of hepatic vein occlusion 30 days after transplantation was compared. There were 180 patients in the defibrotide group, 2 of whom developed hepatic vein occlusion, accounting for 12%; there were a total of 176 patients in the control group, of which 35 developed hepatic vein occlusion, accounting for 20%. The use of defibrotide significantly reduced the incidence of the disease, and the effect was obvious.

The above is the effect of treating hepatic veno-occlusive disease. I hope it will be helpful to you.
