非布索坦治疗痛风疗效好吗?

There are more and more gout patients in modern society. Many patients cannot reduce their uric acid for a long time and are forced to take drugs with serious side effects such as benzbromarone and colchicine. There may even be uric acid crystal deposition, joint pain, and very painful. However, it is the latest drug to treat gout. So, is febuxostat effective in treating gout?

Compared with allopurine drugs, febuxostat only has an inhibitory effect on reduced XOR, while febuxostat has a significant inhibitory effect on both oxidized and reduced XOR, so its effect of reducing uric acid is more powerful and lasting.

Since allopurinol is a purine analog, it will inevitably affect the activities of other enzymes involved in purine and pyridine metabolism. Therefore, repeated high-dose administration is required to maintain high drug levels during allopurinol treatment. This also leads to serious or even fatal adverse reactions due to drug accumulation. Febuxostat is a non-purine XOR inhibitor and therefore has a better safety profile.

In addition, gout is usually treated by promoting uric acid excretion and inhibiting uric acid production, and taking appropriate measures to improve related symptoms.

The production of uric acid in the body is related to purine metabolism. In the final step of purine metabolism, hypoxanthine generates xanthine under the action of xanthine oxidoreductase (XOR), and then further generates uric acid. Inhibiting the activity of this enzyme can effectively reduce the production of uric acid.

Febuxostat inhibits uric acid production by inhibiting xanthine oxidase (XO). Allopurinol can inhibit the reduced form of XO, while febuxostat has a good inhibitory effect on both the reduced and oxidized forms, so its uric acid-lowering effect is more powerful and faster.

Although the interaction between febuxostat and drugs metabolized by XO (such as chophylline, mercaptopurine, azathioprine) has not been studied, the inhibitory effect of this product on XO will increase the concentration of these drugs in the plasma and cause toxicity. It is contraindicated for use by patients taking azathioprine, mercaptopurine or chophylline.
