What is zoledronic acid? Injection (Zoledronic Acid) was approved by the State Food and Drug Administration of China on May 22, 2009, for the treatment of postmenopausal osteoporosis in women.
Osteoporosis and osteoporotic fractures have become one of the major risk factors threatening public health in my country. Osteoporosis is a systemic bone disease in which bone density and bone quality are reduced due to various reasons, and bone microstructure is destroyed, resulting in increased bone fragility and prone to fractures. Postmenopausal osteoporosis generally occurs in women within 5 to 10 years after menopause; senile osteoporosis generally refers to osteoporosis that occurs in elderly people after the age of 70; and idiopathic osteoporosis mainly occurs in adolescents, and the cause is still unknown. Zoledronic acid can be used in patients with osteoporosis. This drug was originally used to treat patients with tumor bone metastasis, but it is also applicable to patients with osteoporosis.
The difference between Zoledronic Acid and other bisphosphonates is that its R2 side chain group is a dinitrogen-containing imidazole ring. This structure enables the drug to firmly bind to hydroxyapatite on the bone surface. In vitro studies have confirmed that compared with other bisphosphonates, zoledronic acid has a stronger binding force to hydroxyapatite on the bone surface and has a stronger inhibitory effect on osteoclasts. Zoledronic acid accumulates in active sites of bone resorption and is taken up by osteoclasts during the bone resorption process. It directly inhibits farnesyl pyrophosphate (FPP) synthase, limiting the synthesis of osteoclast microstructural proteins, causing the cells to lose function and eventually undergo apoptosis.
Medical companions remind: Zoledronic Acid is contraindicated in patients who are allergic to this product or other bisphosphonic acid drugs. It is not recommended for patients with severe renal insufficiency. It is contraindicated in pregnant and lactating women.
