What is Tolvaptan?

Tolvaptan tablets (Tolvaptan) is a selective vasopressin V2 receptor antagonist, used to slow the decline in renal function in patients at risk for rapidly progressive autosomal dominant polycystic kidney disease (ADPKD). It is also approved for the treatment of hypervolemia and hypervolemic hyponatremia, such as congestive heart failure, cirrhosis, and syndrome of inappropriate antidiuretic hormone secretion (SIADH).

Urinary output and fluid intake generally increase in a dose-dependent manner, resulting in an overall negative fluid balance in patients taking tolvaptan. Increases in serum sodium and osmotic pressure can be observed 4-8 hours after administration and are maintained for 24 hours. The magnitude of changes in serum sodium and osmolality increased with increasing dose.
