3. Usage and dosage

1. Recommended dose: The initial recommended dose of leborexan is5 mg once a night, and it is recommended to take it before going to bed. Patients should ensure that they allow at least 7 hours of sleep after taking the medication. Based on the patient's clinical response and tolerability, the dose may be adjusted to the maximum recommended dose of 10 mg, but only once daily.

2. Dosage adjustment

1) When used concomitantly withCYP3A inhibitors or inducers: Avoid coadministration of leborexan with strong or moderate CYP3A inhibitors. If combined use is necessary, the recommended maximum dose is 5 mg.

2) If used in combination with a weakCYP3A inhibitor, the maximum dose should also be 5mg.

3) Leboresen should also be avoided when combined with strong or moderate CYP3A inducers.

4) Patients with hepatic impairment:For patients with moderate hepatic impairment, the maximum recommended dose is5 mg once nightly. Leboresen is not recommended for patients with severe hepatic impairment.

3. Medication recommendations: Leborexen should be taken without or before meals to reduce the delayed impact of the drug on sleep time. Patients should avoid taking medications after or shortly after meals.

4. Adverse reactions

Common adverse effects of leborexan include drowsiness, fatigue, headache, and nightmares or abnormal dreams. In clinical trials, some patients also reported uncommon side effects such as sleep paralysis and hypnotic hallucinations. If the patient experiences any symptoms of discomfort, it is recommended to communicate with the doctor in time to adjust the medication or take necessary treatment measures.

5. Supply and storage

Leborexan tablets are available in two dosage forms: 5mg and 10mg, and patients should use them according to their doctor's prescription. This medication should be stored at room temperature between 20°C and 25°C (68°F and 77°F), with temperature fluctuations between 15°C and 30°C (59°F and 86°F) allowed. Direct sunlight and humid environments should be avoided.

7. Mechanism of action

Leborexan regulates arousal drive in patients with insomnia by antagonizing orexin receptors (OX1R and OX2R). In patients with insomnia, the signaling effects of orexin A and orexin B usually promote wakefulness, and leborexan works by blocking the binding of these neuropeptides to receptors, thereby improving sleep.
