How long does it take for Nintedanib/Vigat to take effect?
Nintedanib (Nintedanib) is an antipulmonary fibrosistargeted drug, and its efficacy usually requires a certain amount of time to accumulate. Unlike traditional anti-inflammatory drugs, nintedanib does not demonstrate its efficacy by rapidly improving symptoms, but by long-term inhibition of fibroblast activity and slowing down the remodeling of lung tissue structure, thereby delaying disease progression. Generally speaking, patients can gradually observe a slowdown in the rate of decline in lung function after taking it for 3 to 6 months. This is also a common evaluation period in international clinical studies.

The main mechanism of action of nintedanib is to block the signaling of vascular endothelial growth factor receptor (VEGFR), platelet-derived growth factor receptor (PDGFR) and fibroblast growth factor receptor (FGFR), thereby inhibiting the abnormal fibrosis process from the root. Because fibrosis is a chronic and accumulated pathological change, it takes time for drugs to work. In the early stages, patients may feel that their symptoms are not significantly improved, but lung function test indicators such as FVC (used to measure changes in lung volume) can usually be observed to slow down significantly after long-term use. Multiple overseas long-term follow-up results show that the efficacy of nintedanib is mainly reflected in "delaying deterioration" rather than "immediate improvement." This means that its real value lies in prolonging the stable period of the disease and preventing further deterioration of respiratory function.
In actual clinical practice, doctors generally recommend that patients continue to take medication for at least half a year, and evaluate the efficacy in combination with imaging, pulmonary function testing and symptom scores. If the patient tolerates it well during the treatment, it can be maintained for a long time. Some patients even continue to take it for several years to control and stabilize their disease. It should be emphasized that if treatment is interrupted in the short term due to adverse reactions such as diarrhea or elevated liver enzymes, it may affect the accumulation of therapeutic effects. Therefore, the continuity of drug use should be maintained through dose reduction or symptomatic and supportive treatment.
