Causes of resistance to Trastuzumab

Trastuzumab resistance (Trastuzumab) is caused by multiple mechanisms. Among them, cell membrane protein expression and abnormal activation of intracellular signaling pathways play an important role. The study discovered another mechanism by which YB-1 causes trastuzumab resistance. By comparing the differences in gene expression and protein expression between sensitive cell lines and drug-resistant cell lines, members of the mitogen-activated protein kinase regulatory protein (MNK) family related to acquired resistance were screened out. The expression of MNK-1 continues to increase in drug-resistant cell lines. Up-regulating or down-regulating MNK-1 expression can significantly affect the sensitivity of cell lines to trastuzumab. Using the drug BI-D1870 to inhibit the activity of MNK-1 kinase RSK can reverse trastuzumab resistance.
