Qlosi eye drops are a prescription drug used to improve near vision in adults with presbyopia.

Precautions for Qlosi Eye Drops

1. General warnings and precautions

(1) Blurred vision: May cause accommodation spasm, resulting in blurred vision, dimness or sensitivity to light. Until your vision is clear again, you should avoid dangerous behaviors that require clear vision, such as driving, operating machinery, or engaging in night activities.

(2) Risk of retinal detachment: Although rare, there have been reports of retinal detachment in susceptible individuals or patients with existing retinopathy using Qlosi eye drops. It is recommended to conduct a retinal examination before taking the medication. If you suddenly experience flashes of light, floaters, or vision loss, seek medical attention immediately.

(3) Use with caution in patients with iritis: It is not recommended for patients with active iritis because it may cause adhesion between the iris and the lens (posterior iris synechiae).

2. Medication for specific groups of people

(1) Pregnant and lactating women: There is currently a lack of relevant clinical data. It should be used only when absolutely necessary after weighing the pros and cons.

(2) Elderly people (65 years and above): Clinical studies have not included a sufficient number of elderly subjects, and its safety and effectiveness have not yet been established. Use with caution.

(3) Children: Presbyopia is an age-related disease and is not suitable for children.

3. Usage and operation details

(1) Contact lens wearers: Remove the lenses before instilling the drops, and wait at least 10 minutes after instilling the drops before wearing them again.

(2) Avoid contamination and eye damage: Avoid contacting the tip of the medicine bottle with the eyes or any surface when instilling eye drops to prevent contamination. Single vials are for single use only and should be discarded after use.

(3) Combined use of other eye drops: If multiple eye drops need to be used, there should be at least a 5-minute interval between different drops.

Qlosi Eye Drops Drug Interactions

QLOSI mainly works locally on the eyes, with very little systemic absorption. Specific drug interaction research results are not clearly listed in the official instructions. Based on its pharmacological mechanism (cholinergic receptor agonist), the following theoretical interaction possibilities need to be considered when using it:

1. Combined use with other cholinergic drugs

Concurrent use of other systemic or topical drugs with cholinergic effects (such as some drugs for the treatment of glaucoma) may enhance the pharmacological effects and adverse reactions of QLOSI (such as blurred vision, sweating, gastrointestinal discomfort, etc.).

2. Combined use with anticholinergic drugs

Anticholinergic drugs (such as atropine, etc.) may antagonize the miosis and accommodation effects of QLOSI, thus affecting its efficacy in improving presbyopia.

3. Important Tips

If you need to use multiple topical eye drops, you must follow the principle of at least 5 minutes interval. This is mainly to ensure that the first drug is fully absorbed and avoid being flushed and diluted by the second drug, so as to ensure the efficacy of each.

Patients should proactively inform their doctors of all the drugs they are taking, including prescription drugs, over-the-counter drugs and health care products, so that the doctor can conduct a comprehensive evaluation.

Pharmacokinetics of Qlosi eye drops

1. Absorption

(1), time to peak (Tmax): After administration on the 8th day, the median time for the drug concentration in the plasma to reach the peak was 2.20 hours.

(2) Peak concentration (Cmax): The average peak plasma concentration after administration on the 8th day was 897.2pg/mL.

(3) Exposure (AUC): The area under the average plasma concentration-time curve after administration on the 8th day was 2699hr*pg/mL.

These data show that although there is a small amount of systemic absorption of QLOSI after eye drops, the blood concentration is extremely low (pg/mL is picogram/ml, which is a very trace unit).

2. Half-life

In the same study, the average elimination half-life of the drug in plasma was measured to be 3.96 hours. This means that the concentration of the drug in the blood circulation decreases by half every approximately 4 hours, indicating that it is cleared from the body more quickly.

3. Elimination

Qlosi eye drops are mainly metabolized by the liver, with an average plasma elimination half-life (T₁/₂) of 3.96 hours. Metabolites are mainly excreted by the kidneys, with no obvious drug accumulation.
