About LeBron

Lemborexant is an OX1/OX2 receptor antagonist of the protein-coupled orexin receptor, which is used to treat insomnia characterized by difficulty falling asleep and/or maintaining sleep. Leborexan tablets were developed and researched by the Japanese pharmaceutical company Eisai. They were approved for marketing in the United States in December 2019 under the trade name Dayvigo. They were subsequently launched in Japan and Canada in July and November 2020 respectively, and in Taiwan, China in September 2021. The dosage form is tablets and is divided into two specifications: 5 mg and 10 mg. Clinical trial results show that Lebraxen can significantly improve the subjective awakening state, shorten the sleep onset latency, and significantly increase the sleep maintenance time.

Indications of Lebraxen

Leborexan is indicated for the treatment of insomnia in adults characterized by difficulty initiating and/or maintaining sleep.

Leborexen usage and dosage

Leborex is 5 mg and should be taken no more than once per night at bedtime, at least 7 hours before scheduled awakening. The dose may be increased based on clinical response and tolerability to the maximum recommended dose of 10 mg. If taken with or shortly after a meal, the time to sleep may be delayed.

Do not increase your dose of leboresen without telling your healthcare provider first. If your insomnia (sleeping problem) worsens or does not improve within 7-10 days, contact your healthcare provider. This could mean there's another condition causing sleep problems.

If you take an overdose of leborexan, contact your healthcare provider immediately or go to the nearest hospital emergency room. You may still feel drowsy the day after taking leborexan. Do not drive a vehicle or engage in other dangerous activities until you feel fully awake.

Leborex drug interactions

1. Taking strong, moderate and weak inhibitors of CYP3A4 at the same time can increase the AUC and Cmax of leborexan, thereby increasing the incidence of adverse drug reactions. Therefore, avoid coadministration of leboresen with strong and moderate inhibitors of CYP3A4, and do not exceed 5 mg of leboresen when coadministered with weak inhibitors of CYP3A4.

2. Taking leborexan together with strong or moderate inducers of CYP3A will reduce its exposure and may lead to a decrease in efficacy. Therefore, avoid taking it with the above two types of drugs.

3. Taking leboresen together with CYP2B6 substrates will reduce the AUC of this type of drug and reduce the efficacy of this type of drug. Therefore, patients who take leboresen and CYP2B6 substrate drugs at the same time need to be closely monitored and consider increasing the dose of CYP2B6 substrate drugs.
