非那雄胺服药注意事项
As a 4-azasteroid compound, finasteride tablets are commonly used in clinical treatment of symptomatic benign prostatic hyperplasia. Finasteride is a specific inhibitor of the intracellular enzyme type II 5α reductase during the metabolism of the androgen testosterone into dihydrotestosterone. Inhibition of this enzyme can hinder the conversion of testosterone into the androgen dihydrotestosterone in peripheral tissues, leading to a significant decrease in the concentration of dihydrotestosterone in serum and tissues.
What are the precautions for taking finasteride?
Genotoxicity: In vitro bacterial and mammalian cell mutation tests and in vitro alkaline elution test results did not show mutagenic effects. In the in vitro CHO cell chromosome aberration study, finasteride slightly increased the CHO cell chromosome aberration rate at a concentration of 450 to 550 μmol. This concentration is equivalent to 4000 to 5000 times the peak plasma concentration after oral administration of 5 mg of this product. In the in vivo chromosomal aberration test, mice were given finasteride 250 mg/kg/day (according to AUC, equivalent to 228 times the recommended human clinical daily dose of 5 mg, the dose calculation method for all toxicological studies below is the same), and there was no increase in chromosomal aberrations.
Reproductive toxicity: 80mg/kg/day (calculated as above, equivalent to 543 times the human daily dose), continuous administration for 12 weeks has no effect on the fertility of sexually mature male rabbits and male rats. When rats were given 80mg/kg/day of finasteride for more than 24 weeks, the weight of their seminal vesicles and prostates was also significantly reduced, and the formation of sperm plugs failed during mating, resulting in decreased fertility in rats; however, it had no effect on the testicles and mating behavior of rats and rabbits; the above toxic effects recovered within 6 weeks after discontinuation of the drug.
