Mechanism of action of TPOXX
The mechanism of action of TPOXX is different from that of cidofovir. It is a nucleoside analogue that can inhibit the replication of viral DNA because it can completely and actively resist cidofovir-resistant vaccinia virus. TPOXX targets a highly conserved virally encoded protein (commonly known as p37) that is present in all orthopoxviruses. In keeping with other viral and cellular proteins, p37's role in the orthopoxvirus replication cycle is to coordinate the formation of folded strip-shaped virions with strong outward expansion while promoting virus spread from infected cells and dissemination in the host. Represents the feasibility and possibility of being used as a drug to treat smallpox and other pathogenic orthopoxvirus diseases.
About smallpox
Smallpox is a highly fatal infectious disease caused by the variola virus. In the 17th and 18th centuries, smallpox caused a global pandemic, killing at least 300 million people worldwide. In human history, plagues such as smallpox, the Black Death, and cholera have left staggering death tolls, and have even been called by historians "the largest genocide in human history" that did not rely on guns.
TPOXX’s mechanism of action
The mechanism of action of TPOXX against smallpox virus is different from that of cidofovir in that it acts on a conserved target unique to poxviruses that is essential for replication. Some scholars have studied the mechanism of action of TPOXX against pox viruses and found that the target of TPOXX is the product of the V061 gene. The vaccinia virus V061 gene is the homologous gene of the vaccinia virus F13L gene. TPOXX can cause changes in the intracellular location of the F13L gene. By changing the cellular location of the F13L gene, it inhibits its activity, thereby inhibiting the assembly and maturation of extracellular viruses and preventing the spread of viruses between cells. This may be the mechanism of action of TPOXX against pox viruses.
