埃索美拉唑治疗胃食管反流性疾病疗效如何?

Esomeprazole (Nexium) is the S-isomer of omeprazole. It reduces gastric acid secretion through a specific targeting mechanism and is a specific inhibitor of proton pumps in parietal cells. How effective is esomeprazole in the treatment of gastroesophageal reflux disease?

A clinical trial was conducted to observe the efficacy of esomeprazole (Nexium) in the treatment of gastroesophageal reflux disease (GERD). The clinical trial randomly divided 94 patients who met the diagnostic criteria for GERD into an esomeprazole treatment group (44 cases) and an omeprazole (OME) control group (50 cases). After 4 to 8 weeks of treatment, the relief of clinical symptoms, changes in gastric pH, and the healing of some patients with esophagitis were observed.

Results: After 4 weeks of treatment, symptoms such as heartburn, chest pain, and acid reflux were significantly improved in the ESO group than in the OME group (P less than 0.05). The intragastric pH value test showed that the esomeprazole group was lower than the OME group (P less than 0.05). Gastroscopy was performed at the 4th and 8th weeks after taking the drug. The healing status of patients with esophagitis in the esomeprazole group was better than that of the patients in the OME group (P<0.05). Conclusion: Esomeprazole (Nexium) is more effective than omeprazole in the treatment of GERD.

Adverse reactions that have been identified in clinical trials of esomeprazole include: Central and peripheral nervous system: paresthesias, drowsiness, insomnia, and dizziness. Reversible confusion, agitation, aggression, depression and hallucinations, mainly in patients with severe disease. Endocrinology: Gynecomastia Gastrointestinal: Stomatitis and gastrointestinal candidiasis Hematology: Leukopenia, thrombocytopenia, pancytopenia. Liver: Encephalopathy (in patients with previous severe liver disease); jaundiced or non-icteric hepatitis; liver failure Musculoskeletal: Arthralgia, muscle weakness and myalgia. Skin: rash, photosensitivity, erythema multiforme, Stevens-Johnson syndrome, etc.
