Dosage and usage of dordaviprone-Modeyso

Dordaviprone (dordaviprone)-Modeyso is a treatment for diffuse midline glioma, specifically indicated for adults and pediatric patients 1 year of age and older who have H3 K27M mutations and have disease progression on prior therapy.

Before administering medication, patient selection must first be made. Doctors need to determine whether a patient is suitable for Modeyso treatment based on the presence of the H3 K27M mutation in the tumor specimen. There are currently no FDA-approved tests available to confirm this mutation, so caution is needed when selecting patients. In addition, patients need to undergo medical monitoring before treatment, including electrocardiogram (ECG) and electrolyte level testing, and perform relevant monitoring regularly during treatment according to clinical needs. This is an important step to ensure patient safety while receiving medication.

For recommended dosage and administration, the recommended dose of Modeyso for adult patients is 625 mg orally once weekly, and patients should continue at this dose until disease progression or unacceptable toxicity. During use, doctors will adjust the treatment plan according to the patient's specific conditions to ensure that the treatment effect is balanced with the patient's tolerance.

For pediatric patients, the dose of Modeyso is individually adjusted based on body weight. The recommended dosage for pediatric patients 1 to 17 years of age who weigh at least 10 kg is as follows: 125 mg orally weekly for patients 10 kg to < 12.5 kg; 250 mg orally weekly for patients 12.5 kg to < 27.5 kg g; 375 mg orally weekly for patients 27.5kg to <42.5kg; 500mg orally weekly for patients 42.5kg to <52.5kg; and 625 mg orally weekly for patients ≥52.5kg. This weight-based dose adjustment can better meet the needs of patients of different ages and weights.

When takingModeyso, patients should take it on an empty stomach, that is, at least 1 hour before or 3 hours after eating. Capsules should be swallowed whole. For patients who are unable to swallow the capsule whole, the capsule can be opened before taking, the contents mixed with approximately 15 to 30 ml of liquid (such as sports drink, apple juice, lemonade or water) and taken orally as a liquid. It should be noted that once mixed, it should be taken within 2 hours. If this time is exceeded, discard and prepare a new dose.
