Introduction: Phenomenate belongs to a class of drugs called anticonvulsants, which work by reducing abnormal electrical activity in the brain. Phenobarate is used alone or with other medications to treat certain types of partial seizures (seizures that involve only one part of the brain) in adults. Phenobarate is a new anti-epileptic drug that provides a new option for the treatment of epilepsy.
Mechanism of action
It has two mechanisms of action. It can block sodium channels, preferentially weaken the sustained sodium current, and enhance GABA activity through positive allosteric regulation of GABA receptors. Phenobarate is a voltage-gated sodium channel (VGSC) blocker that selectively blocks the inactive state of VGSC and preferentially inhibits sustained sodium currents. Phenobarate may also enhance presynaptic release of gamma-aminobutyric acid (GABA), thereby increasing inhibitory GABAergic neurotransmission.
The efficacy of Phenobarate in the treatment of epilepsy
Phenobarnate is a highly effective drug for the treatment of focal epileptic seizures. More than 20% of patients with drug-resistant epilepsy have achieved seizure remission. In clinical studies, Phenobarate has been shown to significantly reduce the frequency of epileptic seizures in patients. For example, in some studies, different doses of phenobarate reduced seizure frequency by 36% (100 mg), 44% (200 mg), and 47% (400 mg) compared with baseline levels.
The pooled estimated hazard ratio for achieving seizure freedom was 3.71 in the phebanate group compared with the placebo group. Additionally, during the maintenance phase, 50.1% of patients randomized to receive phenobarnate and 23.5% of placebo-treated participants experienced at least a 50% reduction in seizure frequency. Adjuvant phenobarnate therapy is associated with a greater reduction in seizure frequency compared with placebo in adult patients with uncontrolled focal seizures.
Phenobarate Side Effects
Phenobarate is FDA-approved because it is safe and effective for most people who take it, but all drugs carry risks, and some side effects caused by phenobarate can be serious or even life-threatening. It is important to be aware of these serious reactions and know their symptoms. The most common side effects of phebanate are drowsiness, dizziness, fatigue, double vision, and headache. Rare but serious side effects include QT interval shortening, suicidal thoughts or behaviors, and neurological reactions. It is recommended that you inform your doctor promptly for treatment and do not drive or operate machinery until you have had enough experience with phenoparate to determine whether it adversely affects your abilities.
