Anamorelin has a good effect on improving appetite. Clinical trials have shown that anamorelin can significantly increase lean body mass index and improve the symptoms of anorexia and cachexia in patients with advanced non-small cell lung cancer, without a significant increase in serious adverse events.

About cancer cachexia

Cancer cachexia is a multifactorial disease, a catabolic syndrome associated with uncontrolled muscle breakdown and characterized by muscle loss leading to significant weight loss. This may be related to fat loss, functional impairment, changes in body composition, and nutritional disorders. The incidence rate is high in advanced tumors and is an indicator of poor prognosis.

Ghrelin receptors are involved in the release of growth hormone (GH) from the pituitary gland and increase appetite via the hypothalamus. Growth hormone secreted by the pituitary gland stimulates the liver to secrete insulin-like growth factor 1 (IGF-1) and promotes muscle protein synthesis. Anamorelin is a ghrelin receptor agonist used to treat cancer cachexia. As an oral mimetic of ghrelin, it promotes growth hormone secretion by activating ghrelin receptors, increases appetite, leads to an increase in muscle mass and weight, and can improve appetite and muscle mass.

Improve appetite

One study evaluated the efficacy and safety of anamorelin in Japanese patients with cancer cachexia with low body mass index (BMI), non-small cell lung cancer or gastrointestinal tumors with cancer cachexia (BMI <20 kg/m2, involuntary weight loss; 2% in the last 6 months, and anorexia). Patients received 100 mg of anamorelin once daily for up to 24 weeks.

Results: A total of 102 patients were included. Age (71.0±8.2) years old, body mass index (17.47±1.48) kg/m2.

Conclusion: Anamorelin can improve body weight and anorexia-related symptoms in patients with low-BMI cancer cachexia, has a long-lasting effect, and is safe and well-tolerated. The study found that anamorelin improved weight and anorexia-related symptoms in these patients, and these effects lasted for up to 24 weeks. In addition, anamorelin is generally well tolerated. These findings suggest that anamorelin may be a valuable treatment option for patients with low body mass index cancer cachexia.
