Erdafitinib is a kinase inhibitor used to treat adult patients with locally advanced or metastatic urothelial carcinoma who meet the following conditions:

(1) Have FGFR3 or FGFR2 gene mutation;

(2) Disease progression during or after at least one previous platinum-based chemotherapy regimen, including within 12 months of treatment with neoadjuvant or adjuvant platinum-based chemotherapy regimens.

Effects of Erdafitinib:

Data from BLC2001, a multicenter, open-label, single-arm trial that enrolled 87 patients. These patients were diagnosed with locally advanced or metastatic urothelial carcinoma that had progressed during or after at least one prior chemotherapy regimen and had certain FGFR3 gene mutations or FGFR2 or FGFR3 gene fusions. Patients received a starting dose of 8 mg once daily, with the dose increased to 9 mg once daily for patients whose serum phosphorus levels fell below the target of 5.5 mg/dL between days 14 and 17. The starting dose was increased to 9 mg once daily in 41% of patients. Use until disease progression or intolerable toxicity. The objective response rate was 32.2%, of which the complete response rate was 2.3% and the partial response rate was 29.9%. The median duration of response was 5.4 months. Responders included patients who had failed prior treatment with anti-PD-L1 or PD-1 drugs, and approximately one-quarter of participants had previously received anti-PD-L1/PD-1 therapy.
